Variation of high molecular weight glutenin subunits in tetraploid wheats of genomic formula AAGG

Abstract:
High molecular weight glutenin subunits present in several accessions ofltetraploid wheats with genomic formula AAGG, T. timopheevi ssp. timopheevi (cultivated) and ssp araraticum (wild), have been analysed by one- and two-dimensional electrophoretic separations and reversed phase high performance liquid chromatography (RP-HPLC). Combination ofthese techniques have permitted to associate different type of subunits to the A or the G genome and assess extent of allelic variation present at corresponding encoding loci. Results have also revealed that in both subspecies accessions are present possessing expressed y-type subunit at the Glu-A1 locus. This is in contrast with what observed in tetraploid wheats of genomic formula AABB where only in the wild species T. turgidum ssp. dicoccoides such subunit is present. Electrophoretic and chromatographic characteristics of x- and y-type subunits, present at the Glu-A 1 locus in both subspecies, are very similar to those observed for x- and y-type subunits present in the wild diploid wheat T. urartu.
