RP-HPLC of glutenins from cultivated and wild wheats

Reversed phase-high performance liquid chromatography ( RP-HPLC ) has been utilized to separate and characterize, in combination with electrophoretical techniques, allelic variants of high molecular weight glutenin subunits in several cultivars of durum and bread wheats and in several wild diploid and tetraploid wheat progenitors. Significant differences in retention times have been observed for x- and y-type subunits encoded by genes present on the A, B or D genomes. Some subunits, which showed identical apparent molecular weight when separated on SDS­ PAGE gels, were found to have different retention times when separated by RP-HPLC.
