The genus Vitis is rich in biodiversity, but grapevine germplasm needs to be preserved to safeguard genetic resources of the genus and to avoid the loss of wild species as well as of neglected cultivars and clones. The most used method is the cultivation in dedicated vineyards, but availability of additional preservation techniques is advisable. Cryopreservation can potentially guarantee safe, long-term storage of grapevine germplasm. The aim of this study was the set up of a cryopreservation protocol for the wine grape cultivar Nebbiolo. Several parameters were studied. Best results were obtained applying an Incapsulation-Vitrification technique to apical tips of young shoots. To test the possibility to eradicate viruses infecting the mother plant through cryotherapy, part of the shoot tips used as explants were obtained from GVA and GLRaV-3 infected plants. Only one plant was regenerated from these explants: the plant resulted virus-free at the RT-PCR assays performed.
