Abstract:
The electrophoretic heterogeneity of storage proteins particularly gliadins, can confer a high level of discrimination among wheat cultivars. This attribute, combined with the stability of the gliadin electrophoregram in response to common environmental factors, gives to the electrophoresis test a relevance in cultivar identification.
Such information may indicate also wheat or flour suitable for specific application used in marketing, certifying, registering, or breeding wheats. Two dimensional separations of gliadins offer improved resolution since overlapping components in the first dimension can be separated in the second dimension allowing to increase the number of resolved components. A methodology to facilitate identification and comparison of Italian durum wheat cultivars based on separation of gliadins in two dimensional procedure is described in this work.
