abstract:
Genes coding far major albumins in Pisurn. Major Albumin (MA) of P. sativum
and P. abyssiuicum are seed proteins composed by either two or one polypeptides
which arc i?nherited as single rnendelian genes, Gcnomic blots of DNA prepared from
CV. Feltharn First, line 993. (P. sativum, line 809 (P. abyssinicum) and from a
mutant showing no major albumins (null albumin mutant) hybridised against a
cDNA for rnajor albumin revealed the presence of a small polimorphic gene family.
When analyzed at high stringency no restriction fragments omologous to the cDNA
probe were detected in the null alburnin mutant, while some restriction fragments
appear at moderately low stringency and the pattern indicate the possible presence
of a large delection in the rnutant. Northern analyses of total RNA from the rnutant
revealed no rnessages for MA proteins either at high or low stringcncy. One of the
genes coding for MA has been isolated forn a genomic library constructed in
EMBL3 and subcloned in either puC18 and M13 for sequence analysis.
