Abstract:
The fungus Spilocaea oleagina causes peacock leaf spot in olive. Virtually
nothing is known about S. oleagina despite the loss of crop yield caused
by this fungus. In order to get insight, an in vitro culture of the fungus
has been established and its identity confirmed by amplified fragment
length polymorphism analysis. Using this in vitro culture, we have cloned
and analysed the DNA sequences of the 18S and 28S ribosomal RNA genes
(rDNA) as well as the internal transcribed spacers (ITS) and 5.8S rDNA
region of S. oleagina. Sequence analysis and comparison to other fungi
determined that this fungus belongs to the Dothideomycetes class. We have
also determined that S. oleagina is an anamorphic phase of a yet
unidentified Venturia species based on phylogenetic analysis using the 28S
rDNA and ITS sequences.
